چکیده

Solventogenic Clostridium species are motile Gram positive anaerobic bacteria used for the fermentative production of acetone-butanol-ethanol (ABE). The addition of CaCO3 to semi-defined P2 medium was investigated to evaluate its impact on growth, amelioration of butanol toxicity, glucose utilization, and ABE production by solventogenic Clostridium species. Two Clostridium species commonly used in industrial production of fermentative-derived ABE were used in the study. Under culture conditions of the present study, CaCO3 enhanced growth of and total ABE production by solventogenic Clostridium species (C. acetobutylicum ATCC 824 and C. beijerinckii 260 produced more than 24 and 22 g/L ABE, respectively) by 31% to 46%. Dramatically increased growth of solventogenic Clostridium species and fermentation rate resulted in greater ABE concentration and productivity for treatments containing CaCO3 relative to the control with no CaCO3 addition. The addition of CaCO3 into P2 medium, in addition, increased butanol tolerance by more than 40% indicating that elevated levels of ABE produced by solventogenic Clostridium species may have occurred due to enhanced tolerance of butanol in the presence of CaCO3.

Proteome Reference Map and Comparative Proteomic Analysis between a Wild Type Clostridium acetobutylicum DSM 1731 and its Mutant with Enhanced Butanol Tolerance and Butanol Yield

The solventogenic bacterium Clostridium acetobutylicum is an important species of the Clostridium community. To develop a fundamental tool that is useful for biological studies of C. acetobutylicum, we established a high resolution proteome reference map for this species. We identified 1206 spots representing 564 different proteins by mass spectrometry, covering approximately 50% of major metab...

Prolonged conversion of n-butyrate to n-butanol with Clostridium saccharoperbutylacetonicum in a two-stage continuous culture with in-situ product removal.

n-Butanol was produced continuously in a two-stage fermentor system with integrated product removal from a co-feed of n-butyric acid and glucose. Glucose was always required as a source of ATP and electrons for the conversion of n-butyrate to n-butanol and for biomass growth; for the latter it also served as a carbon source. The first stage generated metabolically active planktonic cells of Clo...

Evaluation of industrial dairy waste (milk dust powder) for acetone-butanol-ethanol production by solventogenic Clostridium species

Readily available inexpensive substrate with high product yield is the key to restoring acetone-butanol-ethanol (ABE) fermentation to economic competitiveness. Lactose-replete cheese whey tends to favor the production of butanol over acetone. In the current study, we investigated the fermentability of milk dust powder with high lactose content, for ABE production by Clostridium acetobutylicum a...

Strategies for improved isopropanol–butanol production by a Clostridium strain from glucose and hemicellulose through consolidated bioprocessing

BACKGROUND High cost of traditional substrates and formation of by-products (such as acetone and ethanol) in acetone-butanol-ethanol (ABE) fermentation hindered the large-scale production of biobutanol. Here, we comprehensively characterized a newly isolated solventogenic and xylanolytic Clostridium species, which could produce butanol at a high ratio with elimination of ethanol and conversion ...

Enhanced isopropanol–butanol–ethanol mixture production through manipulation of intracellular NAD(P)H level in the recombinant Clostridium acetobutylicum XY16

Background The formation of by-products, mainly acetone in acetone-butanol-ethanol (ABE) fermentation, significantly affects the solvent yield and downstream separation process. In this study, we genetically engineered Clostridium acetobutylicum XY16 isolated by our lab to eliminate acetone production and altered ABE to isopropanol-butanol-ethanol (IBE). Meanwhile, process optimization under pH...
